Research Project Title: Establishing an Antigen Presenting Cell Line for Mechanobiology

The human immune system recognizes and responds accordingly with appropriate immunological mechanisms against foreign antigens that invade the body or diseased cells within the body. Any dysfunction in the immune system leads to a range of medical pathologies. For instance, failure to arrest progressive diseases such as cancer and when the immune system recognizes healthy cells as foreign, it results in the development of autoimmune diseases. Pivotal in launching the appropriate mechanism to ensure immune homeostasis are the antigen presenting cells (APC). While many previous studies have demonstrated the effects of soluble biochemical signals on immune cells, the biomechanical forces that have been shown to govern the biology of many other cell types have been neglected. Here we propose to examine the effects of the biomechanical cues within the microenvironment of APC on their ability to invoke immune responses. In particular, we plan to fully characterize a commercially available monocytic cell line to determine, once differentiated into APC, their ability to serve as a representative cell line for mechanobiology studies. We then plan to identify the effects of substrate stiffness and topography on APC immune response in an effort to better understand the microenvironmental biomechanical influences on APCs.

Responsibilities Of The Position

  • Take a lead role, demonstrate initiative, and assume laboratory responsibilities in the experimental part of the project.
  • Exercise high standards and maintain quality control in experimental planning, documentation, data collection and archiving.
  • Participate in experimental and analytical training workshops and activities in the lab.
  • Work in team environment, participate in laboratory maintenance and write internal reports.
  • Participate in lab meetings through research updates and journal club.
  • Weekly meetings with PI.

Essential Qualifications

BSc in Biology/Immunology/Molecular Biology

Or

BSc in Bioengineering/Engineering with bio-inspirations

Preferred Experience / Skills

MSc in Biology/Immunology/Molecular Biology. Experience in Molecular techniques (PCR)/Cell culture techniques.

Or

MSc in Bioengineering. Experience in cell culture techniques.
